This glycerine-derived "polymeric ketal" can be turned into a polymeric 1,2-diol by acid-catalyzed aqueous cleavage or transketalation with ethylene glycol (C.C.Leznoff and J.Y.Wong). Both polymer derivatives can be reacted with carbonyl compounds. So Fmoc-amino aldehydes and acetals (e.g. Fmoc-NHCH₂CH(OCH₃)₂) bound to this resin yield, after routine Fmoc/tBu SPPS and acid-catalyzed cleavage, useful peptide derivatives, e.g. for ligation. Hirose et al. obtained the chitinase inhibitor argadin containing an Asp β-aldehyde residue via Fmoc-SPPS employing acetalization to the liberated resin-bound glycerine.
